Hi Rose,
You can do it either way. Some people move to Australia on a fiancee visa and get married, other people get married in the States and then apply for a spouse visa. It's up to you.
As for the visa process. I would recommend, first of all, you visit the Australian Immigration website and download Booklet 1: Partner Migration
http://www.immi.gov.au/allforms/booklets/books1.htm
Print it out, if possible, and read it.
Then go to the IARC website and print out the information sheets for fiancee and spouse visas:

Between those two documents, you'll have most of the information you need to make an application. You can print off all the forms from the Immigration website.
There are a few internet forums out there where you can talk to other Americans who have/are going through exactly the same process, and can give you some further insight:
